Mannogalactoglucan from mushrooms protects pancreatic islets via restoring UPR and promotes insulin secretion in T1DM mice

Type 1 diabetes mellitus (T1DM) lacks insulin secretion due to autoimmune deficiency of pancreatic β-cells. Protecting islets and enhancing has been therapeutic approaches. Mannogalactoglucan is the main type polysaccharide from natural mushroom, which potential medicinal prospects. Nevertheless, antidiabetic property mannogalactoglucan in T1DM not fully elucidated. In this study, we obtained neutral fraction alkali-soluble Amillariella mellea polysaccharide (AAMP-N) with structure fruiting body Armillaria mellea and investigated value AAMP-N T1DM. We demonstrated that lowered blood glucose improved symptoms mice. activated UPR signaling pathway maintain ER protein folding homeostasis promote vivo. Besides that, promoted synthesis via upregulating expression transcription factors, increased Ca2+ signals stimulate intracellular secretory vesicle transport activating CamkII cAMP/PKA signals, enhanced fusion plasma membrane VAMP2. Collectively, these studies on function, indicating could be nutraceutical used for treatment
